ABSTRACT

Objetivo: Avaliar a associação da concentração de Cromogranina A com a ansiedade e estresse em profissionais de enfermagem e a associação da ansiedade e estresse com fatores sociodemográficos, epidemiológicos e laborais. Método: Estudo transversal desenvolvido com 210 profissionais de enfermagem de uma instituição hospitalar do Sudeste de Minas Gerais, Brasil, entre 2018/2019, por meio de preenchimento de um questionário de caracterização, do Inventário de Ansiedade de Beck e do Inventário de Sintomas de Stress para Adultos de Lipp, bem como da coleta de saliva dos participantes para verificar a concentração de Cromogranina A dos participantes. Os dados foram analisados de forma descritiva e inferencial, com nível de significância de 5%. Resultados: A maioria dos profissionais apresentou estresse (58,1%) e ansiedade (51,9%). Fatores como faixa etária, número de filhos, tempo de atuação na instituição, carga horária de trabalho, falta de atividade física, uso de medicamentos e outro emprego, aumentaram as chances desses trabalhadores terem ansiedade e estresse em níveis mais altos (P< 0,001). A Cromogranina A apresentou maiores alterações nos profissionais do turno da tarde. Observou-se associação entre o grupo que tinha estresse e ansiedade com a Cromogranina A, no turno da noite, bem como do grupo que tinha apenas ansiedade em horários do turno da manhã (P< 0,05). Conclusão: Os profissionais apresentaram níveis de ansiedade e estresse, que foram associados às alterações da CgA em alguns horários, demonstrando que essa proteína pode ser um possível biomarcador de ansiedade e de estresse.


Objective: To evaluate the association of Chromogranin A concentration with anxiety and stress in nursing professionals and the association of anxiety and stress with sociodemographic, epidemiological, and occupational factors. Materials and Methods: Cross-sectional study carried out with 210 nursing professionals from a hospital in the southeast of the state of Minas Gerais, Brazil, between 2018 and 2019, by completing a characterization questionnaire, the Beck Anxiety Inventory and the Lipp's Stress Symptoms Inventory for Adults, as well as the collection of saliva from the participants to verify the concentration of Chromogranin A. Data were analyzed descriptively and inferentially, with a significance level of 5%. Results: Most of the professionals had stress (58.1%) and anxiety (51.9%). Factors such as age group, number of children, time working in the institution, workload, lack of physical activity, use of medication and having other jobs, increased the likelihood of these workers experiencing higher levels of anxiety and stress (P< 0.001). Chromogranin A showed greater changes among the nursing professionals working the evening shift. A relationship was observed between the group that presented stress and anxiety with Chromogranin A, during the night shift, as well as the group that presented only anxiety during the morning shift (P< 0.05). Conclusion: Nursing professionals showed levels of anxiety and stress that were associated with changes in CgA at certain times, demonstrating that this protein may be a possible biomarker of anxiety and stress.


Objetivo: Evaluar la asociación de la concentración de Cromogranina A con la ansiedad y el estrés en profesionales de enfermería y la asociación de la ansiedad y el estrés con factores sociodemográficos, epidemiológicos y laborales. Método: Estudio transversal desarrollado con 210 profesionales de enfermería de un hospital del Sudeste de Minas Gerais, Brasil, entre 2018/2019, mediante la realización de un cuestionario de caracterización, el Inventario de Ansiedad de Beck y el Inventario de Síntomas de Estrés para Adultos de Lipp, así como la recolección de saliva de los participantes para verificar la concentración de Cromogranina A de los participantes. Los datos fueron analizados de forma descriptiva e inferencial, con un nivel de significancia del 5%. Resultados: La mayoría de los profesionales tenían estrés (58,1%) y ansiedad (51,9%). Factores como grupo de edad, número de hijos, tiempo de trabajo en la institución, carga de trabajo, falta de actividad física, uso de medicamentos y otros empleos, aumentaron las posibilidades de que estos trabajadores tuvieran ansiedad y estrés en niveles más altos (P< 0,001). La Cromogranina A mostró mayores cambios en los profesionales del turno vespertino. Se observó asociación entre el grupo que presentó estrés y ansiedad con Cromogranina A, en el turno de la noche, así como el grupo que presentó únicamente ansiedad en el turno de la mañana (P< 0,05). Conclusión: Los profesionales presentaron niveles de ansiedad y estrés, que se asociaron con cambios en la CgA en determinados momentos, demostrando que esa proteína puede ser un posible biomarcador de ansiedad y estrés.

SUMMARY OBJECTIVE: In this article, we investigated the association of chromogranin A with coronary artery disease. METHODS: Biochemical parameters and chromogranin A levels obtained from peripheral blood samples during coronary angiography were analyzed in 90 patients. Patients were classified into two groups, namely, SYNergy between PCI with TAXUS and Cardiac Surgery score ≥1 (n=45) and SYNergy between PCI with TAXUS and Cardiac Surgery score=0 (n=45). This is a cross-sectional, prospective study. RESULTS: Serum chromogranin A levels were significantly higher in the group with SYNergy between PCI with TAXUS and Cardiac Surgery score ≥1 compared to the group with SYNergy between PCI with TAXUS and Cardiac Surgery score=0 (1381.5±418.9 ng/mL and 1121.2±290.7 ng/mL, respectively; p=0.002). Serum chromogranin A levels were correlated with SYNergy between PCI with TAXUS and Cardiac Surgery score (r=0.556, p<0.04). ROC analysis showed that the area under the curve for serum chromogranin A levels was 0.687 (p=0.007), and the best cutoff value of 1,131 ng/mL had a sensitivity of 67% and a specificity of 65% for the prediction of coronary artery disease. CONCLUSION: Serum chromogranin A levels were increased in coronary artery disease patients with SYNergy between PCI with TAXUS and Cardiac Surgery score ≥1. Increasing serum chromogranin A levels are proportional to the SYNergy between PCI with TAXUS and Cardiac Surgery score.

ABSTRACT

Background:Measurement of biomarkers representing sympathetic tone and the surgical stress response are helpful for objective comparison of anesthetic protocols. Aims: The primary aim was to compare changes in chromogranin A levels following pump pediatric cardiac surgery between children who received bolus caudal morphine and those who received a conventional intravenous narcotic?based anesthesia regime. The secondary objectives were to compare hemodynamic responses to skin incision and the magnitude of the rise in blood sugar values between the groups. Settings and Design: A prospective observational study at a tertiary cardiac center. Measurements and Methods: Sixty pediatric cardiac surgical patients were randomized to Group I [n = 30] to receive intravenous narcotic?based anesthesia and Group II [n = 30] to receive single?shot caudal morphine. Baseline and postoperative chromogranin A levels, the hemodynamic response to skin incision, changes in blood sugar levels, and the total intravenous narcotic dose administered were recorded for each participant. Statistical Analysis: Pearson’s Chi?squared test was used for comparison of categorized variables, and Mann–Whitney test was used for the analysis of continuous data. Results: Changes in chromogranin A levels and blood sugar levels were comparable in both groups. Group II received a lower narcotic dosage (P ? 0.001), and the response to skin incision as reflected by systolic pressure rise was less (P = 0.006). Conclusions: Surgical stress response attenuation was similar to caudal morphine as compared with intravenous narcotic?based anesthesia techniques as reflected by a similar increase in chromogranin A levels

Abstract: There is a lack of evidence on the correlation between salivary biomarkers and subjective measures of dental fear and anxiety in children. This systematic review aimed to retrieve the scientific evidence comparing the results of dental anxiety measured by salivary biomarkers with patient-reported outcomes in pediatric dental setting. The PECOS was as follows: population: pediatric patients aged ≤ 18 years; exposure: patient-reported outcome measures, such as scales and/or questionnaires; comparator: salivary biomarkers; outcome: anxiety, fear, phobia or stress during dental treatment; study design: observational studies or controlled trials. Electronic searches were conducted in PubMed, Scopus, Web of Science, and Ovid databases. Studies that compared scales/questionnaires and salivary biomarkers for the evaluation of dental anxiety, fear, and stress in children/adolescents during dental treatment were included. Certainty of evidence was assessed with GRADE. Risk of bias of the included studies was assessed with the Cochrane tool or the University of Adelaide tool. From the 314 studies identified, eight were included. Participants' age ranged from three to 13 years. The most used salivary biomarkers and instruments were cortisol and the Dental Subscale of the Children's Fear Survey Schedule, respectively. Most studies showed a weak correlation between objective and subjective measures. The main issues regarding bias were on allocation concealment, blinding of assessors, follow up, and exposure assessment. Certainty of evidence was low/very low. Evidence of salivary biomarkers and patient-reported outcome measures to investigate anxiety, fear and stress in children during in the dental environment is limited. There was no correlation between subjective and objective measures in almost all included studies.

ABSTRACT Objective: To determine the association of red cell blood counts, and liver panel tests to predict outcomes in patients with gastroenteropancreatic neuroendocrine tumors who underwent systemic antineoplastic treatments. Methods: Patients with gastroenteropancreatic neuroendocrine tumors in systemic treatment were assessed according to laboratory tests within the same period. Progression free survival was determined by the period between the beginning of treatment and the date of progression. We used conditional models (PWP model) to verify the association between laboratory tests and tumor progression. The level of significance used was 5%. Results: A total of 30 treatments given to 17 patients in the intention-to-treat population were evaluated. Treatment included octreotide, lanreotide, everolimus, lutetium, and chemotherapy. We had statistically significant results in chromogranin A, neutrophils and platelets-to-lymphocyte ratio. The risk of progression increases by 2% with the addition of 100ng/mL of chromogranin A (p=0.034), 4% with the increase of 100 neutrophil units (p=0.006), and 21% with the addition of 10 units in platelets-to-lymphocyte ratio (p=0.002). Conclusion: Chromogranin A, neutrophils and platelets-to-lymphocyte ratio were associated with disease progression during systemic treatment in gastroenteropancreatic neuroendocrine tumors. Further prospective studies with larger cohorts are necessary to validate our findings.

Objetivo: Descrever a presença do polimorfismo da região codante do gene CHGA Glu264Asp e associa-lo com as características clinicas da doença. Método: Trata-se de um estudo transversal, descritivo e de caso controle para investigar o polimorfismo da CHGA Glu264Asp por meio de amostras de sangue de 53 indivíduos sendo o grupo caso, formado por 23 pacientes, de ambos os sexos, que sofreram a tireoidectomia e foram submetidos ao tratamento com o Radiofármaco Iodeto de Sódio (I131). Para a genotipagem foi utilizada a técnica PCRRFLP. Foi adotado o nível de significância de 5%. Resultados: Houve diferença estatística significativa na distribuição alélica entre indivíduos com câncer papilífero da tireoide e os sadios. Com tudo, a presença do alelo G é um fator de risco para o câncer papilífero da tireoide. Observou-se uma correlação entre o genótipo GG com o aumento do nível de TSH em pacientes que apresentavam essa patologia. Conclusão: Demostrou-se que o alelo selvagem pode ser um fator de risco para o desenvolvimento da neoplasia tireoidiana do tipo papilar. Por ser uma doença de etiologia multifatorial, são necessários outros estudos em populações diferentes para melhor compreensão da doença.


Objective: To describe the presence of polymorphism of the codante region of the CHGA Glu264Asp gene and to associate it with the clinical characteristics of the disease. Method: This is a cross-sectional, descriptive and case-control study to investigate chga glu264Asp polymorphism through blood samples from 53 individuals, consisting of 23 patients of both sexes who underwent thyroidectomy and underwent treatment with the Radiopharmaceutical Sodium Iodide (I131). Pcr-RFLP technique was used for genotyping. The significance level of 5% was adopted. Results: There was a statistically significant difference in the allelic distribution between individuals with papillary thyroid cancer and healthy individuals. With everything, the presence of the G alllet is a risk factor for papillary thyroid cancer. A correlation was observed between the GG genotype and the increased level of TSH in patients with this pathology. Conclusion: It was shown that the wild allomay be a risk factor for the development of the thyroid neoplasm of the papilar type. As it is a disease of multifactorial etiology, further studies in different populations are needed to better understand the disease

Método: Estudio transversal, descriptivo y de casos y controles para investigar el polimorfismo chga glu264Asp a través de muestras de sangre de 53 individuos, constituido por 23 pacientes de ambos sexos sometidos a tiroidectomía y tratamiento con yoduro de sodio radiofarmacéutico (I131). Se utilizó la técnica Pcr-RFLP para el genotipado. Se adoptó el nivel de significancia del 5%. Resultados: Hubo una diferencia estadísticamente significativa en la distribución alélica entre individuos con cáncer papilar de tiroides e individuos sanos. Con todo, la presencia del Gallete G es un factor de riesgo para el cáncer papilar de tiroides. Se observó una correlación entre el genotipo GG y el aumento del nivel de TSH en pacientes con esta patología. Conclusión: Se demostró que el aloplasma silvestre puede ser un factor de riesgo para el desarrollo de la neoplasia tiroidea del tipo papilar. Como es una enfermedad de etiología multifactorial, se necesitan más estudios en diferentes poblaciones para comprender mejor la enfermedad

Objective:To explore the clinical significance of serum Chromogranin A (CGA) level in predicting the prognosis of children with severe hand, foot, and mouth disease (HFMD) and complicating neurogenic pulmonary edema (NPE).Methods:A total of 162 patients with HFMD admitted in our hospital from January 2017 to December 2019 were enrolled in the study; and 40 age-matched healthy children were selected as controls. According to the disease severity and complication the patients were divided into three groups: mild group ( n=88), severe without NPE group ( n=46) and severe with NPE group ( n=28). In 72 severe HFMD patients 16 cases died (fatal group) and 56 cases survived (survival group) within 28 days of hospitalization. The serum CGA, LAC, GLU, WBC, PCT, IL-6, cTnT were measured in all subjects. SPSS 23.0 software was used for data analysis, and the receiver operating characteristic (ROC) curve was used to evaluate the various indicators for predicting the prognosis of severe HFMD combined with NPE. Results:The serum CGA, GLU, LAC, IL-6 and cTnT levels in severe HFMD group with NPE significantly higher than those in the other three group ( H=61.554, 79.031, 86.994, 36.477, 75.021, all P<0.05 ). The serum CGA, LAC, GLU and IL-6 levels in the fatal group were significantly higher than those in survival group ( Z=-6.094, -4.621, -4.283, -5.504, all P<0.05). There was no significant difference in the levels of WBC, PCT and cTnT between the survival group and the fatal group ( P>0.05). The area under the receiver operating curve (AUC) of serum CGA was 0.890 (95% CI: 0.833-0.947) for predicting the prognosis of patients and the best cut-off value was 120.59 μg/L. Conclusion:The detection of serum CGA levels may be beneficial for the early diagnosis of severe HFMD with NPE, and can be used as one of the predictors of death from severe HFMD.

Here we report a 25 years old male patient came to our emergency department with chief complaints of headache from 3 months, blurring of vision 7 days, abdominal pain 7 days, and vomiting from 2 days. Young male, with no known comorbidities, presented to our casualty with headache since, 3 months, episodic in nature, increased over the last 1 week, holocranial, gradually progressive in severity. last recorded Blood pressure outside was 220/120 mmHg and papilledema were noted on fundoscopy. USG abdomen and pelvis, CECT abdomen done and diagnosed to have bilateral adrenal pheochromocytoma. Patient underwent bilateral adrenalectomy was performed with good intra op BP control. Right adrenals were enlarged and shows multiple nodular lesions, left adrenal noted shows nodular lesions. Surgery remains the first line of management in malignant pheochromocytoma. Norepinephrine, epinephrine, and dopamine all act on their target receptors, which causes a physiological change in the body. Increase in catecholamines cause severe hypertension and multiple systemic complications like (cardiovascular, cerebrovascular events) and can lead to death if untreated.

ABSTRACT Introduction: Cerebrovascular diseases have been associated with several genes. Chromogranin A (CHGA) has been used as maker in cardiovascular disease. Therefore, evaluating the polymorphism and verifying its association with this pathology is very important to better understand this disease. Objective: The aim of this study was to identify the association between coding region polymorphism in -264 position of the CHGA gene (Glu264Asp) and hemorrhagic stroke (HS)/aneurysm in the Federal District, Brazil. Methods: This is a population-based case-control, involving 45 cases with HS and/or aneurysm. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method is used for genotyping these samples. A significance level of 5% was adopted. Results: The absence of the CC genotype the Glu264Asp CHGA polymorphism in the study participants and the significant presence of the GC heterozygote genotype were observed in this study. However, the distribution of genotypes did not differ statistically in the groups. Conclusion: The Glu264Asp CHGA polymorphism does not seem to contribute to the genesis of the CHGA protein expression in this patients group, but to understand whether or not there is a possible association of the pathology in question and whether the mutation will contribute in the gene therapy and thus to improve patients' quality of life.


RESUMEN Introducción: Enfermedades cerebrovasculares han sido vinculadas a diversos genes. La cromogranina A (CgA) es utilizada como un marcador en enfermedades cardiovasculares. Por consiguiente, evaluar el polimorfismo y verificar la asociación con esa patología es muy importante para la mejor comprensión de la enfermedad. Objetivo: El enfoque del ensayo fue identificar la asociación entre el polimorfismo en la región codificante en la posición -264 del gen CHGA (Glu264Asp) y el accidente cerebrovascular hemorrágico (ACVH)/aneurisma en Distrito Federal, Brasil. Métodos: Estudio de caso-control de base poblacional, involucrando 45 casos con ACVH y/o aneurisma. Para el genotipaje de las muestras, se utilizó la técnica de laboratorio reacción en cadena de la polimerasa-polimorfismos en la longitud de los fragmentos de restricción (PCR-RFLP). Nivel de significación elegido: 5%. Resultados: Ausencia del genotipo CC del polimorfismo Glu264Asp CHGA en los participantes del ensayo y presencia significativa del genotipo heterocigoto GC. Sin embargo, la distribución de los genotipos no difirió estadísticamente en los grupos. Conclusión: El polimorfismo Glu264Asp CHGA parece no contribuir para la génesis de la expresión de la proteína CgA en este grupo de pacientes, pero revelar si existe o no una posible asociación de la patología en cuestión y si la mutación contribuirá para la terapia genética y mejorará la calidad de vida de los pacientes.


RESUMO Introdução: Doenças cerebrovasculares têm sido ligadas a diversos genes. A cromogranina A (CHGA) é utilizada como um marcador em doenças cardiovasculares. Portanto, avaliar o polimorfismo e verificar a associação com essa patologia é muito importante para melhor compreensão dessa doença. Objetivo: O foco do estudo foi identificar a associação entre o polimorfismo na região codante posição -264 do gene CHGA (Glu264Asp) e o acidente vascular encefálico hemorrágico (AVEH)/aneurisma no Distrito Federal, Brasil. Métodos: Estudo caso-controle de base populacional, envolvendo 45 casos com AVEH e/ou aneurisma. Para a genotipagem dessas amostras, utilizou-se a técnica laboratorial reação em cadeia da polimerase-polimorfismo de comprimento de fragmento de limitação (PCR-RFLP). Nível de significância de 5% foi adotado. Resultados: A ausência do genótipo CC do polimorfismo Glu264Asp CHGA nos participantes do estudo e a presença significativa do genótipo heterozigoto GC foram verificadas. No entanto, a distribuição dos genótipos não diferiu estatisticamente nos grupos. Conclusão: O polimorfismo Glu264Asp CHGA parece não contribuir para a gênese da expressão da proteína CHGA nesse grupo de pacientes, mas revela se existe ou não uma possível associação da patologia em questão e se a mutação contribuirá para a terapia gênica e melhorará a qualidade de vida dos pacientes.

Aim: The aim of the study was to compare chromogranin A (CgA) and stress levels before and after non-surgical periodontal therapy (NSPT). Materials and Methods: A sample of 40 patients in the age range 25–60 years were included in the study and were divided into gingivitis (10 subjects), chronic periodontitis (CP) (15 patients) and aggressive periodontitis (AgP) (15 patients). The patients were asked to complete two sets of stress questionnaires, plaque index, gingival index, probing depth and clinical attachment levels (CAL) were recorded. Salivary samples were taken at baseline and were repeated three months post NSPT. Results: CgA was detected in saliva samples of all the groups. A statistically significant correlation was established between levels of CgA and stress parameters, which was shown to be the highest in AgP (P < 0.001), followed by CP group (P < 0.005) at baseline. Following NSPT, an overall reduction was observed in the levels of CgA, which was correlated with the overall reduction in stress levels for AgP group (P < 0.005) followed by CP group (P < 0.037). Amongst the clinical parameters, CAL showed the strongest correlation with CgA both at baseline and after NSPT (P < 0.001). Conclusion: Stress was directly correlated to the levels of salivary CgA levels, which was the highest for aggressive periodontitis at baseline. NSPT showed a marked improvement in all the parameters. Levels of CgA and CAL showed a significant correlation in both the CP and AgP groups.

Introducción: Algunos autores han demostrado incremento de células neuroendócrinas en colitis microscópica y colitis ulcerativa. Objetivo: El objetivo del presente estudio fue evaluar la presencia de células neuroendócrinas en colitis linfocítica, colitis colagenosa y colitis ulcerativa en comparación a controles. Materiales y métodos: Se usó inmunohistoquímica para identificar a las células neuroendócrinas a través del marcador cromogranina A. El estudio incluyó 10 casos de cada diagnóstico de colitis linfocítica, colitis colagenosa y colitis ulcerativa. Resultados: Se encontró diferencia estadísticamente significativa en el conteo de células neuroendocrinas en colitis linfocítica (p=0,019104) y colitis ulcerativa en comparación con los controles (p=0,0077). En colitis colagenosa, se encontró un incremento de células neuroendocrinas pero no pudimos demostrar diferencias estadísticamente significativa. Conclusión: Se demostró hiperplasia de células neuroendocrinas en colitis linfocítica y colitis ulcerativa, lo que confirma lo reportado por los pocos estudios anteriores realizados sobre el tema.


Introduction: Some authors have found increase of neuroendocrine cells in microscopic colitis and ulcerative colitis. Objective: The aim of this study is to evaluate the presence of neuroendocrine cells in ulcerative colitis and lymphocytic colitis and collagenous colitis. Materials and methods: Immunohistochemistry was performed to identify neuroendocrine cells through marker chromogranin A (CgA). The study included 10 cases of each diagnosis of Lymphocytic colitis, collagenous colitis and ulcerative colitis. Results: There was statistically significant difference in the count of neuroendocrine cells, between lymphocytic colitis and control (p=0.019104), and between ulcerative colitis and controls (p=0.0077). In collagenous colitis there was an increase in neuroendocrine cells but we failed to find statistical differences. Conclusion: We could observe neuroendocrine cell hyperplasia in lymphocytic colitis and ulcerative colitis compared with controls, which confirm previous studies.

ABSTRACT

The ganglion cardiacum or juxtaductal body is situated along the left recurrent laryngeal nerve in the aortic window and is an extremely large component of the cardiac nerve plexus. This study was performed to describe the morphologies of the ganglion cardiacum or juxtaductal body in human fetuses and to compare characteristics with intracardiac ganglion. Ganglia were immunostained in specimens from five fetuses of gestational age 12–16 weeks and seven fetuses of gestational age 28–34 weeks. Many ganglion cells in the ganglia were positive for tyrosine hydroxylase (TH; sympathetic nerve marker) and chromogranin A, while a few neurons were positive for neuronal nitric oxide synthase (NOS; parasympathetic nerve marker) or calretinin. Another ganglion at the base of the ascending aorta carried almost the same neuronal populations, whereas a ganglion along the left common cardinal vein contained neurons positive for chromogranin A and NOS but no or few TH-positive neurons, suggesting a site-dependent difference in composite neurons. Mixtures of sympathetic and parasympathetic neurons within a single ganglion are consistent with the morphology of the cranial base and pelvic ganglia. Most of the intracardiac neurons are likely to have a non-adrenergic non-cholinergic phenotype, whereas fewer neurons have a dual cholinergic/noradrenergic phenotype. However, there was no evidence showing that chromogranin A- and/or calretinin-positive cardiac neurons corresponded to these specific phenotypes. The present study suggested that the ganglion cardiacum was composed of a mixture of sympathetic and parasympathetic neurons, which were characterized the site-dependent differences in and near the heart.

ABSTRACT

Background:Epidemiological studies suggest considerable overlap between functional dyspepsia (FD) and irritable bowel syndrome (IBS).Increased duodenal eosinophil counts have been observed in patients with FD,and the density of chromogranin A (CgA)-positive cells was reduced in duodenum in patients with IBS.Aims:To investigate the alterations and significance of duodenal eosinophils and CgA-positive cells in patients with FD and FD-diarrhea-predominant IBS (IBS-D) overlap.Methods:Outpatients with FD and FD-IBS-D overlap who fulfilled Rome Ⅲ criteria from Aug.2014 to Mar.2015 at the First Affiliated Hospital of Fujian Medical University were enrolled.Biopsy specimens were collected from the duodenal bulb for HE staining and CgA immunohistochemical staining.Eosinophils and CgA-positive cells were counted,and their associations with clinical symptoms were analyzed.Results:Twenty-eight FD patients and twelve FD-IBS-D overlap patients were enrolled in this study.Duodenal bulb eosinophil counts were significantly higher in FD-IBS-D overlap patients than in FD patients [(43.83 ± 7.34)/5HPF vs.(22.90 ± 8.93)/5HPF,P < 0.05],while the density of duodenal bulb CgA-positive cells were significantly lower in FD-IBS-D overlap patients than in FD patients [(13.03 ±5.87)/HPF vs.(19.85 ± 9.18)/HPF,P < 0.05].Spearman correlation coefficient analysis revealed that duodenal eosinophil counts were positively correlated with lower gastrointestinal symptoms such as abdominal pain/discomfort and diarrhea,whereas no correlations were found between density of duodenal CgA-positive cells and both upper and lower gastrointestinal symptoms.Conclusions:Duodenal bulb eosinophil counts are increased and CgA-positive cells are reduced in FD-IBS-D overlap patients than in non-overlap FD patients,which may implicate the difference in clinical symptoms between these two functional gastrointestinal disorders.

ABSTRACT

We aimed to investigate the prognostic value of chromogranin A (CgA) in castration-resistant prostate cancer (CRPC). We conducted a systematic literature search of PubMed, Web of Science, and EMBASE for citations published prior to September 2017 that described CgA and CRPC and performed a standard meta-analysis on survival outcomes. Our meta-analysis included eight eligible studies with 686 patients. The results were as follows: progression-free survival (PFS) was associated with CgA level (hazard ratio [HR] = 2.47, 95% confidence interval [CI]: 1.47-4.14, P = 0.0006); PFS was relative to CgA change (HR = 9.22, 95% CI: 3.03-28.05, P < 0.0001); and overall survival (OS) was relative to CgA level (HR = 1.47, 95% CI: 1.15-1.87, P = 0.002). When we divided the patients into two groups according to therapy status, the result for OS relative to CgA level was an HR of 1.26 (95% CI: 1.09-1.45, P = 0.001) in the first-line hormonal therapy group, and an HR of 2.33 (95% CI: 1.40-3.89, P = 0.001) in the second-line hormonal therapy or chemotherapy group. This meta-analysis indicated that a high CgA level had a negative influence on OS and PFS in CRPC patients. In addition, CRPC patients with a rising CgA had a shorter PFS. Further studies are needed to verify the prognostic value of CgA in CRPC.

ABSTRACT

Objective:To prepare the synaptophysin and chromogranin A monoclonal antibodies with clinical evalua -tion.Methods:The fuse gene (Syn and CgA) was designed and it was constructed on the expression vector pET-28a.Then ,the fusion protein was purified.After protein immunization , cell fusion and screening , the target antibodies were selected .Specificity study and correlation coefficient of Syn and CgA was evaluated by clinical sample comparison validation .Results:By screening,two antibodies 3D9 and 4A12,respectively,for Syn and CgA,were obtained.19 kinds of wax block organization were detected by 3D9,4A12 and control antibody(Leica).The statistical results were analyzed ,the results were in good agreement ,and the correlation coefficients were r=0.9892 and r =0.9939, respectively.Conclusion: This method is prepared to obtain the synaptophysin and chromogranin A antibodies successfully and both can be used for immunohistochemistry .This method can also provide some reference for the study of antibody .

ABSTRACT

Objective To investigate the dynamic changes of chromogranin A in patients with pancreatic neuroendocrine neoplasms after laparoscopic surgery,and to monitor the diagnosis and prognosis of patients with real-time monitoring.Methods Forty-five patients with pancreatic neuroendocrine neoplasms were enrolled in this study from March 2013 to March 2015 in First Hospita of Yulin City.All patients were underwent laparoscopic surgery.The surgical results were observed and the levels of postoperative chromogranin A were monitored.Comparisons between groups were evaluated with the x2 test.Follow-up using telephone interview was performed to detect patients' survival and up to January 2017.Results TNM patients with benign and malignant patients in the postoperative chromogranin A examination found that the positive expression rate were 85.0% and 53.0%,the difference between the two groups was statistically significant (P < 0.01) and the corresponding synaptophysine examination were 88.0% and 89.0% (P < 0.05).The positive expression rates of chromogranin A in patients with lymph node metastasis and non-lymph node metastasis were 47.0% and 80.0%,respectively (P < 0.01).The positive expression rates of the corresponding synaptophysin were 87.0% and 87.0%,respectively (P > 0.05).Conclusion Endoscopic surgery for pancreatic neuroendocrine neoplasms is a minimally invasive,safe surgical approach,long-term survival rate of patients with the use of chromogranin A to monitor the level of patients after surgery can effectively assess the development and prognosis of the disease,but the synaptophysine has no significance in assessing the prognosis of the patient.

ABSTRACT

Objective:To investigate the expression level of synaptophysin (Syn), tissue neuronal cell adhesion molecule 56 (CD56) and chromogranin A (CgA) in 92 primary esophageal small cell carcinoma (PESC) and to explore its repationship with clinicopathological features and clinical outcome. Methods:Immunohistochemical studies of CD56, CgA, and Syn were performed in 92 paraffin-embed-ded tissues with clinical-related information obtained from 500,000 esophageal and gastric cardia carcinoma databases established by Henan Key Laboratory for Esophageal Cancer Research of the First Affiliated Hospital of Zhengzhou University in Henan, China. Binary logistic regression was used to analyze the correlations of CgA, Syn, and CD56 expression with clinicopathological features. Kaplan-Mei-er survival analysis and Cox proportional hazards regression models were performed for univariate and multivariate survival analyses. Log-rank test was used to compare the difference in survival rates. Results:The CgA-positive expression rate in PESC at lower segment of esophagus (72.2%) was higher than those at the middle and lower segments (41.1%, 10.0%) (P=0.001). The expression level of CD56, CgA, and Syn was not correlated with gender (P=0.262, 0.998, 0.931), age (P=0.250, 0.998, 0.703), tumor invasion (P=0.253, 0.997, 0.061), and lymph node metastasis (P=0.767, 0.998, 0.613). Univariate analysis showed no survival influence in patients with and without lymph node metastasis (P=0.563). Multivariate survival analysis showed that patients with PESC mixed squamous cell car-cinoma (HR=2.58;95%Cl, 1.11-5.98) and higher CgA protein expression (HR=1.87;95%Cl, 1.02-3.43) exhibited a longer survival time than those with pure PESC and without CgA expression. Conclusion:Tissue CgA level was associated with tumor location in PESC. His-tological type and tissue CgA expression were independent important prognostic factors, and lymph node metastasis exerted no influ-ence on survival in PESC.

ABSTRACT

Objective To observe the influence of Simvastatin on immature rabbit model of chronic heart failure(CHF),and to explore the possible protective mechanism of Simvastatin for the rabbits with CHF.Methods Thirty immature male rabbits were divided into 3 groups randomly:control group,heart failure group (HF group)and Simvastatin group(SIM group),10 rabbits in each group.The models of CHF were established by injecting Adrinmycin via the auricular vein of rabbits (1.5 mg/kg,once 1 week,for 12 weeks).The control group were injected the same amount of 9 g/L saline.SIM group were given both injection of Adrinmycin and Simvastatin [1.5 mg/(kg · d)for 12 weeks].The immature rabbit's cardiac function and myocardial morphology changes were evaluated.The expressions of chromogranin A (CgA) and apoptosis signal-regulating kinase 1 (ASK1) were evaluated.Results (1) In control group,the immature rabbits were all alive;in the other 2 groups,most immature rabbits were depressed and sluggish.The survival rate of HF group was 60%,while in SIM group the survival rate was 80%.(2)Compared with the control group,the left ventricular ejection fraction in HF group and SIM group decreased significantly [(40.05 ± 6.74)%,(50.18 ± 5.73) % vs.(65.93 ± 5.65) %,all P < 0.01],while in SIM group was higher than that of HF group,and the difference was significant (P < 0.05);compared with HF group,the left ventricular end diastolic diameter and left ventricular end systolic diameter decreased in SIM group [(13.48 ± 1.24) mm vs.(16.23 ± 2.82) mm;(9.87 ± 0.85) mm vs.(11.13 ± 1.21) mm],and the differences were significant (all P < 0.05).(3) Compared with the control group,the expression of CgA (mean optical density 142.24 ± 17.14,127.93 ± 12.12 vs.78.65 ± 6.78,P < 0.05;integrated optical density 1 422.41 ± 167.34,1 279.37 ± 118.15 vs.786.54 ± 75.84,P < 0.05) and ASK1 (mean optical density 140.32 ± 18.65,115.48 ± 12.30 vs.69.85 ± 6.54,P < 0.05;integrated optical density 1 403.23 ± 165.67,1 158.79 ± 137.81 vs.698.58 ± 64.51,P < 0.05) increased in the HF group and SIM group.While the expression in the SIM group decreased significantly compared with that of the HF group,and the difference was significant (P < 0.05).Conclusions Simvastatin can improve cardiac function of immature rabbits with CHF.The mechanism of SIM may depress the expressions of CgA and ASK1.

ABSTRACT

Objective To investigate whether catestatin (bovine chromogranin A 344-364) can inhibit the biofilm formation of Candida albicans and examine its relationship with the expression of adhesion gene HWP1.Methods Clinical strains and standard strain ATCC 10231 of C.albicans were studied.XTT [2,3-bis-(2-methoxy-4-nitro-5-sulfophenyl)-2H-tetrazolium-5-carboxanilide] method was used to assess the ability of C.albicans biofilm formation.Antifungal activity against planktonic Candida ceils was evaluated in terms of minimum inhibitory concentrations (MICs) according to the description in CLSI-M27-A3.XTT assay and colony count were used to assess the effect of catestatin on inhibiting C.albicans biofilm formation.The lowest concentration showing 50 % inhibition on biofilm formation (BIC50) was decided by calculating the metabolic activity.The adhesion of C.albicans reduced by catestatin was visualized under an inverted microscope and quantified by colony count.The expression of HWP1 was analyzed by RT-PCR.One-way analysis of variance (ANOVA) and Dunnett's T3 test were used to compare the results.Results Clinical strains and standard strain ATCC 10231 of C.albicans showed strong ability in forming biofilm.Catestatin exhibited MICs ranging from 40 μmol/L to 80 μmol/L against planktonic C.albicans cells,and BIC50 of 80-160 μmol/L in inhibiting C.albicans biofilm formation.Catestatin reduced the adhesion of C.albicans.The colony forming unit (CFU) was 27 822.22-±-2 472.74 in blank control group,while the CFU was 5 355.55± 1 264.03,11 377.78±2 232.58,17 488.89±1 136.27,22 377.78±3 521.99,and 26 044.44±1 329.57 in the presence of 160,80,40,20,and 10 μmol/L catestatin,respectively (F=147.018,P=0.001).The difference between control group and 160,80,and 40 μmol/L catestatin was statistically significant (P<0.05).RT-PCR found the expression of HWP1 in the presence of 160 μmol/L catestatin was about 12.24% of that in blank control group.Conclusions Catestatin can effectively prevent C.albicans biofilm formation.This effect may be related to the down-regulated expression of adhesion gene HWP1 by catestatin,which results in reduced adhesion of C.albicans.Promising clinical prospect is expected for this finding.

ABSTRACT

Objective To investigate the potential roles of chromogranin A in pathogenesis of asthmatic inflammation,and to assess the regulation of montelukast on chromogranin A expression.Methods The rat asthma model was established with ovalbumin,and they were allocated to three groups,named asthma group,control group and montelukast group.The expressions of chromogranin A protein and mRNA at lung tissue were detected by immunohistochemisty or real-time PCR methods,and positive expression intensity of chromogranin A protein was assayed by optical density.The correlation between chromogranin A protein and mRNA was also analyzed.Results The expression levels of chromogranin A protein in asthma group(0.34 ±0.05 optical density)was significantly higher than that in control group (0.21 ±0.06 optical density)(P<0.01 ).The expression levels of chromogranin A mRNA in asthma group (4.02 ±0.95 relative quantity value)was significantly higher than that in control group(P<0.01 ).The expression levels of chromogranin A protein in montelukast group(0.28 ±0.04 optical density)was dramatically lower than that in asthma group (0.34 ±0.05 optical density)(P<0.05),while there were no statistical significance of chromogranin A mRNA(3.67 ±0.78 relative quantity value)between those two groups(P>0.05 ).But levels of mRNA was positively correlated with protein of chromogranin A (r=0.635,P<0.01).Conclusion Expressions of chromogranin A protein and mRNA at lung tissue were increased in asthmatic rats,and the results demonstrated that chromogranin A perhaps participated in the pathogenesis of asthma inflammation,but this function of chromogranin A protein could be down regulated by montelukast.
